Clinical Context

Mantle cell lymphoma is a subtype of non-Hodgkin lymphoma characterized by a poor prognosis and a need for effective treatment strategies. Historically, treatment options have included chemotherapy, immunotherapy, and targeted therapies such as ibrutinib. However, despite advancements, many patients experience relapse or refractory disease. The introduction of zanubrutinib, a selective Bruton's tyrosine kinase (BTK) inhibitor, has shown promise in improving outcomes for patients with relapsed or refractory MCL. Sonrotoclax, a BCL-2 inhibitor, further expands the therapeutic landscape, potentially offering a novel mechanism of action for patients who have exhausted other options. The integration of these agents into the treatment algorithm reflects ongoing efforts to enhance patient outcomes in MCL management.
